RESUMO

A strain high-performance of esterase producing bacteria was screened from soil, which could selectively hydrolyze D-homoserine lactone from its racemate to achieve the resolution of L- homoserine lactone with more than 99% e.e. in 48% yield. L-homoserine lactone building block was then converted to L-α-amino-γ-bromobutyronic acid chiral blocks, which reacted with various nucleophilic reagent modules could to be applied to prepare L-γ- substituted α-amino acids such as L-selenomethionine, L-methionine, L-glufosinate and L-selenocystine. Its advantages included high selectivity of biocatalytic resolution reactions, high optical purity of products, racemic recycle of D-substrates and modular reaction, which simplified the production process of these products and highlighted the power of biological manufacturing.

The Soil Moisture Ocean Salinity (SMOS) was the first mission providing L-band multi-angular brightness temperature (TB) at the global scale. However, radio frequency interferences (RFI) and aliasing effects degrade, when present SMOS TBs, and thus affect the retrieval of land parameters. To alleviate this, a refined SMOS multi-angular TB dataset was generated based on a two-step regression approach. This approach smooths the TBs and reconstructs data at the incidence angle with large TB uncertainties. Compared with Centre Aval de Traitement des Données SMOS (CATDS) TB product, this dataset shows a better relationship with the Soil Moisture Active Passive (SMAP) TB and enhanced correlation with in-situ measured soil moisture. This RFI-suppressed SMOS TB dataset, spanning more than a decade (since 2010), is expected to provide opportunities for better retrieval of land parameters and scientific applications.

Livestock wastewater has led to serious eco-environmental issues. To effectively treat livestock wastewater and realize the resource utilization of livestock solid waste, manure waste has been widely used to prepare biochar for the recovery of nitrogen and phosphorus. However, fresh biochar has a poor ability to adsorb phosphate due to its negative charge. To overcome the defect, the proportion of biochar samples prepared at 400 °C and 700 °C was optimized under a mass ratio of 2:3 to obtain mixed biochar PM 4-7, achieving the purpose of enhanced ammonium and phosphate recovery in livestock wastewater simultaneously without any modification. The effects of pyrolysis temperature, dosage, and pH were studied, different adsorption models were used to explore the adsorption mechanism, and the effect of biochar loaded with nutrient elements on seed was verified through a seed germination experiment. It was revealed that the maximum removal rates of phosphate and ammonium were 33.88 % and 41.50 %, respectively, endorsing that mixed biochar PM 4-7 can recover nutrients from livestock wastewater, and could be used as a slow-release fertilizer to promote seed germination and growth. This method provides a new potential way for the efficient resource utilization of pig manure and the recovery of nutrients from breeding wastewater.

Plants are sessile and encounter to abiotic environmental stressors, such as nutrient deficiency and drought stress. Identifying stress tolerance genes and their mechanisms is vital to ensuring plant survival. In this study, we characterized NCED3 in the tobacco plant Nicotiana tabacum, a key enzyme in the biosynthesis of abscisic acid that is widely involved in abiotic stress responses, using overexpression and RNA interference knockdown. Overexpression of NtNCED3 promoted primary root development, leading to increased dry weight, root-to-shoot ratio, photosynthetic capacity, and acid phosphatase activity, coinciding with highly increased phosphate uptake capability under low phosphate conditions. Under both drought and extreme phosphate deficiency conditions, the phosphate starvation response preceded the drought stress response. However, under high phosphate conditions, the drought stress phenotype emerged before the symptoms of phosphate deficiency. Plants overexpressing NtNCED3 grew better than the wild-type and NtNCED3 knockdown plants, with more developed root systems and higher biomass, phosphorus content, and hormone content. This study provides evidence that NtNCED3 enzyme participates in plant responses to phosphate deficiency and drought stress in N. tabacum, and NtNCED3 may serve as a potentially valuable gene for genetic modification of plant tolerance to both drought stress and phosphate starvation.

Non-specific lipid transfer proteins (nsLTPs) play an important role in plant growth and stress resistance; however, their function in tobacco remains poorly understood. Therefore, to explore the function of NtLTP in response to high temperature, we identified an NtLTPI.38 from tobacco, obtained its overexpression and knockout transgenic plants, and further studied their response to heat stress (42 °C). The results showed that NtLTPI.38 overexpression in tobacco reduced chlorophyll degradation, alleviated the high temperature damage to photosynthetic organs, and enhanced the photosynthetic capacity of tobacco under heat stress. NtLTPI.38 overexpression in heat-stressed tobacco increased the contents of soluble sugar and protein, proline, and flavonoid substances, reduced the relative conductivity, and decreased H2O2, O2•-, and MDA accumulation, and increased the enzymatic antioxidant activities, such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX), compared to wild type (WT) and knockout mutant plants. RT-PCR confirmed that the expression levels of antioxidant enzymes and thermal stress-related genes were significantly upregulated under thermal stress in overexpression plants. Therefore, NtLTPI.38 enhanced heat tolerance in tobacco by mitigating photosynthetic damage and improving osmoregulation and antioxidant capacity. These results provided the theoretical basis and a potential resource for further breeding projects to improve heat tolerance in plants.

Non-specific lipid transfer proteins (nsLTPs) typically have conserved structural resemblance, low sequence identity, and broad biological functions in plant growth and stress resistance. Here, a plasma membrane-localized nsLTP, NtLTPI.38, was identified in tobacco plants. Multi-omics integrated analysis revealed that NtLTPI.38 overexpression or knock out significantly changed glycerophospholipid and glycerolipid metabolism pathways. NtLTPI.38 overexpression remarkably increased phosphatidylcholine, phosphatidylethanolamine, triacylglycerol, and flavonoid levels, but decreased ceramides compared to wild type and mutant lines. Differentially expressed genes were associated with lipid metabolite and flavonoid synthesis. Many genes related to Ca2+ channels, abscisic acid (ABA) signal transduction, and ion transport pathways were upregulated in overexpressing plants. NtLTPI.38 overexpression in salt-stressed tobacco triggered a Ca2+ and K+ influx in leaves, increased the contents of chlorophyll, proline, flavonoids, and osmotic tolerance, and raised enzymatic antioxidant activities as well as the expression level of related genes. However, mutants accumulated more O2- and H2O2, exhibited ionic imbalance, gathered excess Na+, Cl-, and malondialdehyde, with more severe ion leakage. Therefore, NtLTPI.38 enhanced salt tolerance in tobacco by regulating lipid and flavonoid synthesis, antioxidant activity, ion homeostasis, and ABA signaling pathways.

Surface soil moisture (SSM) is an important variable in drought monitoring, floods predicting, weather forecasting, etc. and plays a critical role in water and heat exchanges between land and atmosphere. SSM products from L-band observations, such as the Soil Moisture Active Passive (SMAP) Mission, have proven to be optimal global estimations. Although X-band has a lower sensitivity to soil moisture than that of L-band, Chinese FengYun-3 series satellites (FY-3A/B/C/D) have provided sustainable and daily multiple SSM products from X-band since 2008. This research developed a new global SSM product (NNsm-FY) from FY-3B MWRI from 2010 to 2019, transferred high accuracy of SMAP L-band to FY-3B X-band. The NNsm-FY shows good agreement with in-situ observations and SMAP product and has a higher accuracy than that of official FY-3B product. With this new dataset, Chinese FY-3 satellites may play a larger role and provide opportunities of sustainable and longer-term soil moisture data record for hydrological study.

Eye movements are one of the most fundamental behaviors during reading. A growing number of Chinese reading studies have used eye-tracking techniques in the last two decades. The accumulated data provide a rich resource that can reflect the complex cognitive mechanisms underlying Chinese reading. This article reports a database of eye-movement measures of words during Chinese sentence reading. The database contains nine eye-movement measures of 8,551 Chinese words obtained from 1,718 participants across 57 Chinese sentence reading experiments. All data were collected in the same experimental environment and from homogenous participants, using the same protocols and parameters. This database enables researchers to test their theoretical or computational hypotheses concerning Chinese reading efficiently using a large number of words. The database can also indicate the processing difficulty of Chinese words during text reading, thus providing a way to control or manipulate the difficulty level of Chinese texts.

Using the Visual World Paradigm, the current study aimed to explore whether the mass/count distinction is determined by syntax in Mandarin Chinese, focusing on classified nouns in nominal phrases. By using dual-role classifiers, ontological count and mass nouns, and phrase structures with and without biased syntactic cues we found that the mass/count distinction is initially computed using phrase structure but can be overridden in cases where the syntax is incompatible with nouns' ontological meanings. The results indicate that in Mandarin Chinese, syntactic cues can be rapidly used to make predictions about upcoming information in real time processing.

INTRODUCTION: Mesenchymal stem cells (MSCs) are an excellent donor graft source due to their potential for self-renewal and multidirectional differentiation. However, the potential mechanisms involved in MSC homing and neural differentiation are still unclear. The purpose of this study was to explore the effects of a chemokine, SDF-1a, and Wnt3a ligand on rat MSCs' migration and b-mercaptoethanol (BME)-induced neural differentiation of MSCs. MATERIALS AND METHODS: MSCs were isolated from rat bone marrow and cultured in vitro to passage 3. Scratch tests and transwell assays were used to estimate the effects of SDF-1a (25 ng/mL) and Wnt3a (10 ng/mL) on the migration of MSCs. The expression of Wnt/PCP pathway proteins RhoA, c-Jun, ATF2, and Wnt3a were assessed by Western blot. The 5 mM BME-induced neural differentiation of MSCs was determined by immunofluorescence to detect neuron- and astrocyte-specific markers such as nestin, GFAP, and Olig2. RESULTS: Wnt3a promoted the migration ability of MSCs and regulated the expression of RhoA, c-Jun, and ATF2 proteins. MSCs could differentiate into neural stem cells and astrocytes. Wnt3a enhanced BME induced neurogenesis in MSCs by increasing the protein expression of RhoA, c-Jun, and Wnt3a. CONCLUSIONS: The present study demonstrated that the Wnt/PCP pathway promotes migration and neural differentiation of rat MSC.

Previous research has demonstrated effects of both orthographic neighborhood size and neighbor frequency in word recognition in Chinese. A large neighborhood-where neighborhood size is defined by the number of words that differ from a target word by a single character-appears to facilitate word recognition, while the presence of a higher-frequency neighbor has an inhibitory effect. The present study investigated modulation of these effects by a word's predictability in context. In two eye-movement experiments, the predictability of a target word in each sentence was manipulated. Target words differed in their neighborhood size (Experiment 1) and in whether they had a higher-frequency neighbor (Experiment 2). The study replicated the previously observed effects of neighborhood size and neighbor frequency when the target word was unpredictable, but in both experiments neighborhood effects were absent when the target was predictable. These results suggest that when a word is preactivated by context, the activation of its neighbors may be diminished to such an extent that these neighbors do not effectively compete for selection.

In the current study, we conducted 2 eye-tracking reading experiments to explore whether sentence context can influence neighbor effects in word recognition during Chinese reading. Chinese readers read sentences in which the targets' orthographic neighbors were either plausible or implausible with the pretarget context. The results revealed that the neighbor effect was influenced by context: The context in the biased condition (where only targets but not neighbors can fit in the pretarget context) evoked a significantly weaker inhibitory neighbor effect than in the neutral condition (where both targets and neighbors can fit in the pretarget context). Retinoblastoma (RB) is an intraocular malignant tumor that often occurs in children. Along with the improvement of treatment strategies, the cure rate of RB has increased significantly. However, the treatment of advanced and recurrent RB remains as a critical challenge. Therefore, studying the molecular mechanisms underlying the progression of RB is essential for the development of novel and effective therapeutic strategies. Through the analysis of a previously published microarray study, we found that ribonucleotide reductase subunit M2 (RRM2) was highly expressed in RB tissues as compared to normal tissues. The purpose of this study is to clarify the role and mechanism of RRM2 in regulating the progression of RB. We first demonstrated that RRM2 expression level in RB tissues and cell lines was significantly higher when compared to that in normal retinal tissue and cell lines, and high RRM2 expression level was associated with a poorer overall survival of patients. In RB cells, RRM2 overexpression promoted cell proliferation, migration, invasion and epithelial-mesenchymal transformation (EMT), while RRM2 silencing suppressed these biological features. Silencing RRM2 reduced the activation of Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) signaling pathway, and the presence of JAK2/STAT3 signaling pathway inhibitor INCB attenuated the effect of RRM2 overexpression. Collectively, our data indicate that RRM2 promotes the progression of RB by activating JAK2/STAT3 signaling pathway. Targeting RRM2/JAK2/STAT3 axis lays a theoretical foundation for the formulation of novel RB therapy.

Mesenchymal stem cells (MSCs) are considered a promising tool for treating cerebral ischemic injury. However, their poor survival after transplantation limits their therapeutic effect and applications. Salidroside has been reported to exert potent cytoprotective and neuroprotective effects. This study aimed to investigate whether salidroside could improve MSC survival under hypoxic-ischemic conditions and, subsequently, alleviate cerebral ischemic injury in a rat model. MSCs were pretreated by salidroside under hypoxic-ischemic conditions. The cell proliferation, migratory capacity, and apoptosis were evaluated by means of Cell Counting Kit-8, transwell assay, and flow cytometry. MSCs pretreated with salidroside were transplanted into the rats subsequent to middle cerebral artery occlusion. The grip strength, 2,3,5-triphenyltetrazolium chloride, and hematoxylin-eosin staining were used to analyze the therapeutic efficiency and pathological changes. The mature neuron marker NeuN and astrocyte marker GFAP in the focal area were detected by immunofluorescence. These results indicated that salidroside promoted the proliferation, migration and reduced apoptosis of MSCs under hypoxic-ischemic conditions. In vivo experiments revealed that transplantation of salidroside-pretreated MSCs strengthened the therapeutic efficiency by enhancing neurogenesis and inhibiting neuroinflammation in the hippocampal CA1 area after ischemia. Our results suggest that pretreatment with salidroside could be an effective strategy to enhance the cell survival rate and the therapeutic effect of MSCs in treating cerebral ischemic injury.

AIMS/INTRODUCTION: Emerging evidence has indicated that long non-coding ribonucleic acids play important roles in the development and progression of diabetic retinopathy (DR). It is reported that urothelial carcinoma-associated 1 (UCA1) is highly expressed in diabetic lymphoendothelial cells and influences glucose metabolism in rats with DR. The aim of the present study was to explore the role of UCA1 in the mechanism of DR. MATERIALS AND METHODS: Gene expression analyses in fibrovascular membranes excised from patients with DR using public microarray datasets (GSE60436). Reverse transcription polymerase chain reaction was carried out to detect UCA1, micro-ribonucleic acid (miR)-624-3p and vascular endothelial growth factor C (VEGF-C) expressions in the blood of patients and human retinal endothelial cells (HRECs). Furthermore, Cell Counting kit-8, Transwell assay, and tube formation assay were used to identify biological effects of UCA1 on HRECs proliferation, migration ability and angiogenesis in vitro. RESULTS: UCA1 and VEGF-C were elevated in DR patients and high glucose-induced HRECs cell lines, whereas miR-624-3p was decreased. UCA1 inhibition inhibited proliferation, angiogenesis and migration of HRECs cells under high-glucose condition. Luciferase reporter assay showed that UCA1 could sponge with miR-624-3p, which could directly target VEGF-C. Finally, we proved a pathway that UCA1 promoted cell proliferation, migration and angiogenesis through sponging with miR-624-3p, thereby upregulating VEGF-C in high-glucose-induced HRECs. CONCLUSIONS: We identified UCA1 as an important factor associated with DR, which could regulate the expression of VEGF-C by sponging miR-624-3p in human retinal endothelial cells. Our results pave the way for further studies on diagnostic and therapeutic studies related to UCA1 in DR patients.

Long term surface soil moisture (SSM) data with stable and consistent quality are critical for global environment and climate change monitoring. L band radiometers onboard the recently launched Soil Moisture Active Passive (SMAP) Mission can provide the state-of-the-art accuracy SSM, while Advanced Microwave Scanning Radiometer for EOS (AMSR-E) and AMSR2 series provide long term observational records of multi-frequency radiometers (C, X, and K bands). This study transfers the merits of SMAP to AMSR-E/2, and develops a global daily SSM dataset (named as NNsm) with stable and consistent quality at a 36 km resolution (2002-2019). The NNsm can reproduce the SMAP SSM accurately, with a global Root Mean Square Error (RMSE) of 0.029 m3/m3. NNsm also compares well with in situ SSM observations, and outperforms AMSR-E/2 standard SSM products from JAXA and LPRM. This global observation-driven dataset spans nearly two decades at present, and is extendable through the ongoing AMSR2 and upcoming AMSR3 missions for long-term studies of climate extremes, trends, and decadal variability.

RESUMO

Neurodegenerative disease is a kind of chronic, progressive nervous system disease characterized by neuron degeneration or apoptosis. Current treatments cannot prevent the development of the disease. Possible alternative treatments include cell therapy, especially with the use of mesenchymal stem cells (MSCs). MSCs are pluripotent stem cells with capacities for self-renewal and multidirectional differentiation. MSCs may serve as a reliable source of neural cells for potential cell replacement therapy or regenerative medicine treatment. Here, we summarized the therapeutic mechanisms of MSCs and how they can contribute to the development of treatments for neurodegenerative diseases.

Mesenchymal stem cells (MSCs) are pluripotent stem cells with self-renewal and multidirectional differentiation capabilities. Dimethyloxalyl glycine (DMOG) mobilizes MSCs, and the hypoxia inducible factor-1 (HIF-1) signaling pathway plays an important role in MSC mobilization. We aimed to investigate the effect of DMOG on the HIF-1 pathway in MSCs. Rats were treated with DMOG, and the numbers of peripheral blood MSCs (PB-MSCs) and bone marrow MSCs (BM-MSCs) were detected by the Colony-forming unit fibroblastic (CFU-F) method. The growth curve, cell cycle and migration ability of PB-MSCs and BM-MSCs were detected by CCK-8, Flow cytometry and Transwell assays. Western blotting and real-time qPCR were used to detect the expression of the HIF-1 pathway. The number of bone marrow microvessels was detected by immunohistochemistry. DMOG significantly increased the numbers of PB-MSCs and BM-MSCs (P < 0.05). Further, the MSCs in peripheral blood and bone marrow still had the ability to proliferate and migrate after mobilization by DMOG. The expression levels of HIF-1α, stromal cell-derived factor-1α (SDF-1α) and vascular endothelial growth factor (VEGF) in MSCs were significantly regulated by DMOG (P < 0.05). The number of bone marrow microvessels decreased after the VEGF/VEGFR signaling pathway was blocked by SU5416 (P < 0.05). Therefore, these findings demonstrated that DMOG regulates the HIF-1α signaling pathway and promotes biological effects in MSCs.

Lead-free halide double perovskites (HDPs) are promising candidates for high-performance solar cells because of their environmentally-friendly property and chemical stability in air. The power conversion efficiency of HDPs-based solar cells needs to be further improved before their commercialization in the market. It requires a thoughtful understanding of the correlation between their specific structure and property. Here, the structural and optical properties of an important HDP-based (NH4)2SeBr6 are investigated under high pressure. A dramatic piezochromism is found with the increase in pressure. Optical absorption spectra reveal the pressure-induced red-shift in bandgap with two distinct anomalies at 6.57 and 11.18 GPa, and the energy tunability reaches 360 meV within 20.02 GPa. Combined with structural characterizations, Raman and infrared spectra, and theoretical calculations using density functional theory, results reveal that, the first anomaly is caused by the formation of a Br-Br bond among the [SeBr6]2- octahedra, and the latter is attributed to a cubic-to-tetragonal phase transition. These results provide a clear correlation between the chemical bonding and optical properties of (NH4)2SeBr6. It is believed that the proposed strategy paves the way to optimize the optoelectronic properties of HDPs and further stimulate the development of next-generation clear energy based on HDPs solar cells.

AIM: To investigate the effect of high concentration of glucose (HCG) on double stranded RNA-activated protein kinase-like ER kinase (PERK)-eukaryotic initiation factor-2α (eIF2α)-transcription factor C/EBP homologous protein (CHOP)-cysteine aspartate specific proteinase (caspase-12) signaling pathway activation and apoptosis in rabbit corneal epithelial cells (RCECs). METHODS: RCECs were treated by different concentrations of glucose for 0-48h. The expressions of PERK, p-PERK, eIF2α, p-eIF2α, 78 kDa glucose-regulated protein 78 (GRP78), CHOP, B-cell lymphoma 2 (Bcl-2), B-cell lymphoma-2-associated X protein (Bax) and caspase-12 were determined by Western blot. Apoptosis was detected by TUNEL assay. Meanwhile, the function of PERK-eIF2α-CHOP-caspase-12 signaling pathway activation in high glucose-induced apoptosis was evaluated using PERK inhibitor, GSK2606414. RESULTS: HCG significantly promoted the expression of p-PERK, p-eIF2α, GRP78, CHOP, Bax and cleaved caspase-12 in RCECs (P<0.05), while remarkably decreased the expression of Bcl-2 and caspase-12 (P<0.05), and the alterations caused by glucose were in concentration- and time-dependent manners. Meanwhile, PERK and eIF2α expressions were not affected in all groups (P>0.05). TUNEL assay showed that the apoptosis rate of RCECs in the HCG group increased significantly in contrast with that in the normal concentration of glucose or osmotic pressure control group (P<0.05), and the apoptosis rate increased with the increase of glucose concentration within limits (P<0.05). GSK2606414 down-regulated the expression of p-PERK and p-eIF2α in the HCG group (P<0.05), while still did not affect the expression of PERK and eIF2α among groups (P>0.05). Correspondingly, GSK2606414 also significantly reduced the apoptosis rate induced by high glucose (P<0.05). CONCLUSION: HCG activates PERK-eIF2α-CHOP-caspase-12 signaling pathway and promotes apoptosis of RCECs.
